INFANT Mass Spectrometry Facility

The INFANT MS core facility is engaged in the development and application of mass spectrometry methods for clinical omics and biomarker discovery workflows, with applications in proteomics, metabolomics, and lipidomics.

Instrumentation: Our current MS platforms include the Waters SYNAPT G2-S, a high resolution quadrupole-time of flight (QTOF) instrument and the Waters Xevo TQMS (triple quadrupole).  Both MS instruments are coupled to a Waters Acquity Ultra Performance Liquid Chromatography system.  The SYNAPT offers high resolution/ accurate mass (HR/AM) mass spectrometry for the identification of metabolite/lipid profiles and discovery biomarker profiling.  The Xevo TQMS is a highly sensitive instrument for targeted quantification of analytes, facilitating hypothesis-driven and biomarker validation workflows. We have in-house sample preparation protocols for discovery metabolomics and lipidomics, designed to achieve maximum analyte coverage.  In addition, we have a positive pressure manifold for rapid purification of extracts with solid phase extraction in a 96 well plate format. 

Bioinformatics Software: MassLynx software controls and processes results for both instruments.  In addition, Progenesis Qi software is capable of processing large data files produced by the SYNAPT, it performs peak alignment, normalization, linked database searching and identification of differential features amongst 1000s of compounds.  TargetLynx software is designed for precise calibration and quantification of targeted compounds, and automated batch processing and quality assessment may be performed through the tracking of quality control samples.
